GLP-1 medications and medullary thyroid cancer (MTC) risk
GLP-1 medications carry a boxed warning for medullary thyroid carcinoma based on rodent studies. No human cases have been attributed to GLP-1 use despite 10+ years of post-approval surveillance. Personal or family history of MTC or MEN-2 is a hard contraindication.

How it happens
Rodent thyroid C-cells express GLP-1 receptors at higher density than human C-cells. In long-duration rodent studies, GLP-1 agonists caused C-cell proliferation and medullary thyroid tumors. Human C-cells appear less sensitive — extensive pharmacovigilance data over 10+ years has not detected an MTC signal in humans.

- 1.Tell your prescriber about any personal or family history of thyroid cancer before starting
